Can I sync files located on the D: drive to Dropbox?

Dropbox will only sync the files and folders located in the local Dropbox folder. It can't sync files from other drives or locations.
If you're referring to the Backup feature, your key folders need to be in the original locations for Dropbox to be able to back them up.

Thanks. Why does the app say it can backup files on a removable disk? Isn't it similar to saving data from the second disk D:?

Do you mean the Dropbox app or the Boxifier app? If you mean the Dropbox app, it can backup your external/removable disk to Dropbox. If you mean the Boxifier app, it can sync (not just backup) folders from a local drive (for example your second disk D:), from an external/removable disk or from a network share/drive.
